Novosibirsk Scientists Build a 3D Model of DNA Repair

Scientists in Novosibirsk have, for the first time, obtained a three-dimensional model of how DNA interacts with two enzymes during repair. On October 21, Nikita Bulgakov, a researcher at the Institute of Chemical Biology and Fundamental Medicine of the Siberian Branch of the Russian Academy of Sciences, reported the work. He said researchers had previously relied mainly on calculations and computer predictions.

DNA stores hereditary information in cells. It can be damaged by special molecules that arise during ordinary processes in the body. To repair the damage, cells use enzymes—proteins that speed up chemical reactions.

The work examined DNA and two enzymes, hOGG1 and hAPEX1. According to the study’s description, the first finds the damaged spot and removes part of the DNA. The second helps separate the DNA from the first enzyme and cuts out the rest of the damaged section. The experimental model was built using data from crystals made of DNA and proteins.

The crystallography experiment was conducted at the Sirius synchrotron in the Brazilian city of Campinas. Specialists from the Moscow Institute of Physics and Technology took part. The scientists hope the new data will help explain how cells repair DNA. This may be useful for cancer research, but it does not yet mean a new treatment is available.
